Punicalagin Alleviates Oxidative Stress and Pathological Changes in Brain of Mice-Fed High Fat, High Fructose Diet

The present study investigated to know whether the pomegranate polyphenol punicalagin (PU) can ameliorate oxidative stress and pathological alterations induced by a calorie rich diet comprised of fat fructose. Male Swiss albino mice were fed normal pellet (Control) or high fat-high fructose (HFFD) for 60 days. PU (30 mg/kg bw) was administered orally last 15 days both dietary groups. Measurement TBARS, AOPP FRAP in plasma, histopathology liver brain, assessment damage brain tissues, blood barrier function carried out. HFFD showed an increase levels TBARS AOPP, decline FRAP. administration reduced improved significantly. Histopathology results fatty changes inflammatory infiltration mice. Cortex hippocampus confirmed presence degenerating neurons with shrunken cytoplasm pyknotic nuclei. treatment caused significant hepato neuroprotective effects, among two doses dose (60 mg/kg) more pronounced effects than low mg/kg). immunohistochemical localization markers proteins (3-NT), lipids (4-HNE) DNA (8-OHG) increased tissues upon these significantly decreased. Our findings clearly suggest that modulate could be potential candidate treating neurodegenerative processes associated excess.
